Personally I prefer to use extra substrates like cork granules. Some people use coco soil. You have to be a bit careful with some reptile substrates as they're intended to be moist for reptiles, and too much moistness in a hamster enclosure can lead to mould. There are places that sell coco soil specifically suitable for Hamsters. I'll link some below. Moss - if you decide to use some, you really only need a very small amount of and again it needs to be something suitable for hamsters. It's not really necessary but some people use it to stuff gaps in bendy stick bridges, or a small amount on top of a house eg.
Even with the coco soil linked below, it's not intended to fill an enclosure, but just an additional substrate for a dig box
